Molecular Characterization and Anticancer Activity of Laccase from Fusant Strain of Pleurotus Ostreatus against HuH7 Hepatocarcinoma Cell Line

Abstract

Laccases are blue multi-copper oxidases and catalyze the oxidation of phenolic and non-phenolic compounds. The goal of this study was to characterize the fusant strain of Pleurotus ostreatus at molecular level by Maldi of sequencing method and its ability to act against HuH7 hepatocarcinoma cell lines. In this study the fusant strain was screened for laccase production and it was purified by DEAE- Cellulose chromatographic method. The purified strain was subjected to enzyme activity analysis by UV- Spectrophotometer which shows 0.000115U/ml. It was further estimated for the amount of protein by Lowry's method and it was 0.4mg/ml. The particular fraction was reported for anticancer activity against HuH7 hepatocarcinoma cell lines by MTT assay. The strain used for the anticancer activity shows 97% activity. The results emphasize the promising module to develop a new drug for tumor by using Pleurotus ostreatus with potential strain improvement techniques.
